Orla P Barry

Biography

Dr. Órla P. Barry graduated with an Honours Degree in Biochemistry in 1991 from University College Cork (UCC). This was followed by a Ph.D. at the University of Pennsylvania (UPENN), Philadelphia, USA on "Transcellular activation of platelet microparticles: A role for platelet microparticle arachidonic acid". A post-doctoral fellowship also at UPENN, involved investigating novel mechanisms of transcellular lipid metabolism and isoprostanes as indices of oxidant stress. Upon returning to Ireland at the end of 1999, she joined the Cork Cancer Research Centre as a Health Research Board (HRB) funded Post-Doctoral fellow. Dr. Barry was appointed Lecturer in Pharmacology, UCC, in 2002. She completed the Certificate in Teaching and Learning in 2012 followed by a Diploma in 2013 and graduated with an MA (1H) in Teaching and Learning, in Higher Education, in 2014 through UCC CIRTL (Centre for Integration of Research, Teaching and Learning).

Research Interests

Cell Signalling and Cancer 
Dr. Barry's research focuses on cancers with few treatment strategies and with no significant improvement in overall survival rate, namely, oesophageal and renal cancers. The paucity of therapeutic options, and their associated morbidity, has led to vital research for new therapeutic strategies. Research focuses on the study of different families of serine/threonine kinases that play key roles in signalling mechanisms following activation of seven-transmembrane and tyrosine-kinase receptors. In particular, our area of research focuses on the role of Paks, p38 MAPKs, and PKCs which exert a variety of cellular effects including changes in proliferation, malignant transformation, cell death and differentiation. Briefly, we have exploited the activation of PKC as a pro-apoptotic and anti-proliferative agent in oesophageal cancer. In addition we have re-introduced p38delta MAPK into different cancers which lack this isoform including oesophageal, renal, prostate, lung and liver which has profound effects on cell proliferation and apoptosis. Finally, we have identified different Pak isoforms which are pro-apoptotic in oesophageal and renal cancer. An important aspect of our research is to elucidate the signalling pathways underlying the apoptotic effect of these kinases using both pharmacological and molecular strategies. In addition, the use of athymic nude mice to study their effects is necessary to translate in vitro results to in vivo models of tumourigenesis in order to validate our experimental hypothesis in a physiological setting.  The identification of different kinase isoforms involved in cell proliferation and cell death in different types of cancer may have important therapeutic implications for the treatment of the disease.

Epigenetics and Cancer
Research on epigenetic gene silencing and on epigenetic drug-induced re-expression of tumour suppressor genes may influence future optimal treatment strategies and predict responses in oesophageal and renal cancer patients. Our research identified that epigenetic alterations are partly responsible for the promotion of tumourigenesis in oesophageal squamous cell carcinoma (OESCC). Given that epigenetic changes are reversible, we can investigate the utility of epigenetic modifiers - DNA methyltransferase inhibitors (DNMTi), histone deacetylase inhibitors (HDACi) and Bromodomain and Extra-Terminal motif inhibitors (BETi) as possible therapeutic strategies in the treatment of drug resistant OESCC.  Our work to-date has uniquely identified that some FDA cancer approved and investigational epigenetic modifiers (not all) can prevent the growth of drug resistant p38δ MAPK negative OESCC cell growth.  

Cardiovascular Research 
Cardiovascular research involved investigating the biological roles of platelet microparticles (PMP) formed from the plasma membrane and released into the extracellular space. Prior to my investigations their physiological role(s) remained ill-defined. My research highlighted a novel mechanism of transcellular lipid metabolism whereby platelet activation may be amplified or modulated by concentrated delivery of PMP arachidonic acid (AA) to adjacent platelets, endothelial cells, and monocytes. The demonstration that PMPs may modulate important aspects of endothelial and monocyte function provided a novel mechanism by which platelets may interact with such cells in human atherosclerosis and inflammation. In addition to my cellular and molecular studies of AA signalling and gene transcription, I have also studied the roles of isoprostanes (iPs) as indices of oxidant stress in patients with alcohol-induced hepatitis or alcohol-induced chronic liver disease. iPs are free radical-catalyzed products of AA formed in a COX-independent manner and have proved useful markers for lipid peroxidation.

Research Training
The aim within my laboratory is to train future investigators in an intellectually exciting and supportive work environment to carry out independent, high quality research. Our goal is to educate undergraduate, PhDs and post-doctoral fellows to a level where they have the research knowledge, understanding, critical analytical skills, and the know how to apply appropriate experimental design and state-of-the-art techniques to whatever emerging questions they are attempting to address. In summary, we concentrate on developing analytical, technical and interpersonal skills.
Our laboratory strives to train research scientists in cancer cell and molecular biology focusing on the pharmacological principles of how drugs work and why some are toxic. Our research is conducted at the molecular, cellular, organ, whole organism, and clinical levels. Thus, through individual research driven projects within the department in conjunction with national and international collaborative groups we provide abundant educational opportunities in the broader area of Pharmacology for both undergraduate and postgraduate students.

Teaching & Learning Research (SoTL/TFU/UDL/SDGs)

I have been involved in SoTL research since commencement of my lecturing post in AY0203. Since completing my MA in Higher Education (AY1314) I have been afforded the opportunity to research my teaching and learning (T&L) and in turn student learning and assessment using (i) a teaching for understanding (TFU) lens, (ii) the inclusion of a universal design for learning (UDL) and a scholarship of T&L (SoTL) approach and finally (iii) mappng my teaching to Sustainable Development Goals (SDGs) in particular SDG3 (Good Health and Well-Being), SDG4 (Quality Education), SGD5 (Gender Equality) and SDG12 (Responsible Consumption and Production). I apply the same stringent systematic investigation and methodologies to student learning and assessment as I would to my scientific research. This approach has led to T&L and assessment research collaborations with colleagues in CoMH and CIRTL with success in publishing our research findings and making our teaching and methods of student learning public.
