My research interests lie in the areas of resistance to apoptosis in colon cancer and the molecular mechanisms linking chronic inflammation and tumorigenesis. Within these two areas I have a specific interest in the transcription factors and transcription factor networks which determine the appropriate cellular response to immune¿mediated stress and downstream cell fate decisions. One of the model systems I work with is an in vitro cell culture system, whereby the Th1 cytokine IFN- g is able to sensitize the colon cancer cell line HT29 to TNF- a -triggered apoptosis. One of the intriguing things about this model is that neither IFN- g nor TNF- a alone can trigger apoptosis, only the combination of both cytokines can induce cell death. I have adopted a functional genomics approach using Affymetrix GeneChip expression arrays, siRNA and various small molecule inhibitors to identify the genes, transcription factors and signalling pathways mediating the cell death decision. To date, the IKK b /NF- k B and Wnt/ b -Catenin signalling systems appear to be the major determinants of cell fate in this model.

Another related project I am working on is the identification of novel target genes regulated by PPAR- d . PPAR- d is a member of the nuclear hormone receptor family and is a ligand-activated transcription factor. It is over-expressed in colon cancer and is thought to be a direct b -catenin/Tcf-4 target gene. Recent reports have indicated that COX-2 derived prostaglandin (PGE2) can promote intestinal adenoma formation through PPAR- d , possibly through PPAR- d -mediated suppression of apoptosis. However, there is some controversy in the area and this stems in part from gaps in knowledge regarding PPAR- d function in intestinal epithelial cells. One of these gaps is the lack of information regarding the identity of PPAR- d target genes. In an attempt to address this, I am currently using isogenic cancer cell lines ¾ wild type and null for PPAR- d expression ¾ combined with whole genome Affymetrix GeneChips to identify novel PPAR- d target gene sets in intestinal epithelial cells. The findings from this work may shed some light on the function of this transcription factor in colon cancer and its potential suitability as a drug target.
