André Toulouse

Biography

André Toulouse received a BSc degree in Biology from Université Laval (Québec, Canada) in 1991. He developed his expertise in cellular and molecular biology of diseases by completing a MSc (1993) and a PhD (1998) in Molecular Biology at Université de Montréal (Montréal, Canada). His work characterized the role of retinoic acid receptors in the suppression of lung cancers. His interest in Neuroscience led him to undertake post-doctoral work with Dr Guy Rouleau in the Centre for Research in Neuroscience at McGill University (Montréal, Canada) where he also worked as a research associate in the Neurogenetics Laboratory. His research areas included the identification and cloning of genes involved in neurodevelopmental disorders and the development of cellular models of neurodegeneration for polyglutamine disorders. In 2003, he moved to University College Cork to pursue post-doctoral work on the molecular biology of neurotrophic factors.  

Dr Toulouse was appointed as College Lecturer in Clinical Anatomy and Principal Investigator in the Department of Anatomy and Neuroscience in September 2005. He has taught a vast array of topics related to anatomy and neuroscience and is currently teaching topographical anatomy, human development and molecular neuroscience to students in medicine, dentistry, clinical therapies and neuroscience. In 2008, he completed a post-graduate certificate in Teaching and Learning in Higher Education (UCC) followed by a post-graduate diploma in the same discipline in 2009.  

His research is focused on the development of cellular models for the study of the molecular mechanisms of neurodegeneration in polyglutamine disorders and the role of retinoic acid in brain cancer biology. Through his work as an educator, Dr Toulouse has also developed an expertise in the creation and assessment of teaching tools in neuroanatomy and dental anatomy. In addition, Dr Toulouse is the Director of the Biosciences Imaging Centre, an advanced microscopy facility, at University College Cork. His work has been funded through personal fellowships from the Fonds pour la Formation des Chercheurs et l'Aide à la Recherche and the Fonds de la Recherche en Santé du Québec as well as research grants from the Health Research Board of Ireland, Science Foundation Ireland, the National Forum for Teaching and Learning, and the Irish Research Council for Science, Engineering and Technology.

Research Interests

The role of retinoic acid signalling in brain tumours.
Retinoic acid, a derivative of vitamin A, is a potent signalling molecule in the development of the nervous system. It is responsible for the spatial determination of specific regions of the brain and spinal cord and the terminal differentiation of precursor cells into neural tissue. Studies have shown that specific gene defects in its signalling pathways participate in the development of tumours in a number of tissues, including lung, oesophagus, colorectal and blood. Using a combination of bioinformatics and cell culture models, we are studying the role of various members of the retinoic acid signalling pathway in the development of brain tumours. The identification of specific proteins involved in cancer development allows us to use targeted synthetic drugs to efficiently reduce the growth of cells in vitro.  

Development of cellular models for the study of neurodegeneration and neuroprotection.
The exact nature of cell death mechanisms remains unclear for a large proportion of neurological disorders. Despite the identification of causative genes and chemical agents, the reason why certain neurons are more sensitive to insult than others remains elusive. In addition, despite intensive research efforts, symptomatic treatments often have limited efficiency and cannot prevent further degeneration and progression of the disease. Neuroprotective strategies are being developed, which combined with symptomatic treatment could improve the disease outcomes for patients. Using cell culture models, we replicate important steps in the neurodegenerative process, manipulate them, using drugs and neuroprotective factors, and aim to identify the genes that are responsible for neuronal cell death or that could be used to prevent this process.   Mechanisms of neurodegeneration in CAG trinucleotide repeat expansion disorders The polyglutamine disorders are a group of neurodegenerative disorders caused by the expansion of a coding CAG trinucleotide repeat and include some of the autosomal dominant spinocerebellar ataxias and Huntington’s disease. Although the causative proteins are widely expressed, these disorders are characterized by the degeneration of specific neuronal populations. Several hypotheses have been proposed to explain degeneration, including transcriptional impairment, altered protein interactions, direct polyglutamine toxicity and accumulation of toxic protein by-products following the alteration of the protein degradation pathways but no unifying mechanism has emerged yet.  We have identified a new mechanism of cell toxicity in these disorders where aberrant decoding through ribosomal frameshifting results in the production of highly toxic polyalanine-containing peptides. Recent studies suggest that this mechanism may be involved in many disorders and may be caused by RNA secondary structures, depletion of specific amino acids and the cell’s metabolic rate. Our current studies aim at refining these findings and to further study the mechanism by which the ribosome changes its translational frame. We are developing a cellular model that will allow us to manipulate the translational shifting and explore how cell toxicity can be modulated, identifying potential therapeutic strategies.

Anatomy and Education Research.
Over the past two decades, the number of teaching hours dedicated to the anatomical sciences has been massively reduced in the health sciences curricula, consequently, students are reporting difficulties with the discipline. In addition, limitations in preparing and retaining appropriate anatomical models have further contributed to the increased perception that anatomy is a difficult subject. In recent years, we have developed and assessed a number of educational tools (animations, online resources, 3D computer quiz) to supplement teaching and consolidate learning in the areas of dental anatomy and neuroanatomy. In collaboration with the dissection room technical team, we are interested in exploring new ways of maximizing the use of cadaveric material for teaching and have uncovered a number of rare anatomical variations.

Teaching Activities

Teaching Interests

Undergraduate Teaching
Over the years, I have taught a wide variety of subjects (systematic and topographical anatomy, embryology and developmental anatomy, molecular biology, genetics, molecular and cellular neuroscience, research methods, neuroanatomy) to students from various disciplines (dental hygiene, dentistry, medicine, nursing, clinical therapies, sport studies and education, pharmacy, biological sciences and neuroscience). In addition to developing and updating lecture content, I have established new modules for the B.Ed. in Sports Studies and Education (AN1063) and student selected modules for the medical degree (AN2062 and AN3000). I have re-designed all the modules that I
 currently coordinate, teach and examine (AN1004, AN1006, AN2006, AN2007,  AN2008, AN2060, AN2061, AN2062, AN3000,  AN3009, and AN3011). In addition, I contribute to teaching and examination on the following modules: AN3013, AN4010,  FM1010, FM1020, FM1030, FM1040, FM2102, MH1010, MH1020, MH1030, MH1040, MH2102, GM1002 and MX1009.
 
Undergraduate research projects
I have surpervised a large number of undergraduate research projects and literature reviews from a variety of disciplines including the B.Sc. Neuroscience literature review (AN3011) and research projects (AN4010), B.Sc. Genetics literature review (GN3002) and research projects (GN4002), as well as medical degree research projects (MX1006 and MX5090).
Post-graduate teaching
At post-graduate level, I contribute teaching to the M.Sc in Molecular Cell Biology with Bioinnovation (ML6002) and the M.Sc in Obstetrics and Gynaecology (OG6001 and OG6005). In addition, I deliver the content for module AN6012 - Human Embryology to students in the MSc Human Anatomy programme.
