Ruth Massey

Biography

Biography
Professor Ruth Massey has a B.A. in Natural Sciences and a Ph.D. in Molecular Microbiology from Trinity College Dublin. She worked as a postdoctoral researcher in the UK at both the University of Oxford and Bath, and it was in Oxford where her interest in Gram positive bacterial pathogen began. Ruth’s first independent position was in the Zoology Dept. in Oxford, where Wellcome Trust funding allowed her to set up her first lab, which subsequently moved to Bath in 2007. In 2017 Ruth moved to the University of Bristol and in 2019 was awarded a Wellcome Trust Investigator Award and promoted to Professor of Microbial Pathogenicity. In 2021 Ruth became the SALI Professor for Microbiome and Health Sciences at UCC.

Research Interests

Research Interests
Infectious diseases are among the greatest causes of morbidity and mortality globally. With a dwindling pool of effective antibiotics, pharmaceutical companies shutting down their antibiotic development facilities, and many pathogens remaining recalcitrant to effective vaccine development, new and innovative approaches to infection management and treatment regimens are desperately needed. But without a greater understanding of how bacteria cause disease, we are largely working in the dark, and addressing this has been the focus of Ruth’s research career.

Understanding the interconnectivity between bacterial virulence and antibiotic resistance. There is a delicate balance to be reached by bacterial pathogens between being highly virulent or fully antibiotic resistant, as they are rarely able to be both due to the energetic costs associated both bacterial strategies. Work by Ruth and her team uncovered much of this for the major human pathogen Staphylococcus aureus, where detailed molecular analysis revealed the in-built redundancies and interconnectivity between these strategies that have contributed to the design of clinical trials of potential anti-virulence strategies.

Developing the use of genome sequence data to define bacterial phenotypes and predict patient outcome following infection.
One of Ruth’s major scientific contributions to date began as a concept that molecular and computational approaches could be used to study bacterial phenotypes at a population level, through the development of novel functional genomics approaches. Her team’s first demonstration of the utility of these functional genomics approach with empirical data was published two years later where they successfully predicted the toxicity of individual MRSA isolates from their genome sequence, a seminal paper that has been recommended by Faculty 1000 (https://f1000.com/prime/718345576). To date Ruth’s team’s research approach has facilitated the identification over a dozen novel pathogenicity affecting loci. As they characterise the molecular detail of how each of these loci affect the ability of S. aureus to produce toxins, they are uncovering hidden depths to the complexity that is involved and identifying novel pathways for future therapeutic intervention.

Development of functional genomics approaches to predict patient outcome following infection.
Since Ruth’s early publications in the arena, her research team and have been building upon the complexity of what they are trying to understand about bacterial pathogens. By including information on the source of the infection, they have uncovered hidden subtleties about specific conditions such as the development of and outcome following S. aureus bacteraemia. With the inclusion of patient meta-data they have also been successful in both predicting individual patient mortality, but also identifying several novel features associated with this, including several previously uncharacterised S. aureus proteins. Her most recent work has applied this approach to COVID-19 patient samples demonstrating its utility to combine AMR, host immunity as well as bacterial virulence to understand disease development.
